用BrdU标记技术观察大鼠脊髓上胸段灰质细胞的分化发育 |

【摘要】 目的 观察脊髓灰质细胞的分化发育,寻找脊髓移植时胎鼠脊髓取材的最佳时间。 方法 用5-溴-2-脱氧尿嘧啶(BrdU)标记DNA,ABC免疫细胞化学方法观察大鼠上胸段脊髓灰质细胞的增殖分化情况。 结果孕鼠于E10d腹腔注射BrdU,胎鼠脊髓上胸段BrdU免疫反应神经元主要集中在脊髓后角,脊髓侧角的BrdU免疫反应神经元较少,前角仅有少量的BrdU免疫反应神经元;孕鼠于E11d腹腔注射BrdU,胎鼠脊髓上胸段后角BrdU免疫反应神经元较E10d减少,侧角和前角BrdU免疫反应神经元增多;孕鼠于E12d腹腔注射BrdU,胎鼠脊髓上胸段BrdU免疫反应神经元主要集中于前角和侧角,而后角仅有少量的BrdU免疫反应神经元;孕鼠于E13d、E14d腹腔注射BrdU,胎鼠脊髓上胸段未见BrdU免疫反应神经元。 结论 BrdU标记技术能较好的标记脊髓灰质细胞,胎鼠脊髓进行BrdU标记的最佳时间应选择E12d,已标记胎鼠脊髓的最佳移植取材时间为E13d。 【关键词】 5-溴-2-脱氧尿嘧啶(BrdU);脊髓;发育;移植;大鼠 【中图分类号】 R322.81 【文献标识码】 A 【文章编号】 0529-1356(2000)04-289
APPLICATION OF BrdU LABEL TECHNIQUE FOR STUDYING DIFFERENTIATION AND DEVELOPMENT OF SUPERIOR THORACIC GRAY MATTER OF RAT SPINAL CORD

【Abstract】 Objective To search for appropriate time of embryonic spinal grafting,we have observed the differentiation and development of cells of spinal gray matter. Methods The development and proliferation of spinal gray matter cells of superior thoracic spinal cord were observed by using 5-bromo-2-deoxyuridine(BrdU) to mark DNA and ABC immunocytochemical technique. Results When pregnant rat at E10d was given BrdU by intraperitoneal injection,most of BrdU immunoreactive neurons were located in posterior horn of superior thoracic segment of spinal cord in embronic rat,only a few of BrdU immunoreactive neurons appeared in anterior horn and lateral horn.Given BrdU at E11d,the number of BrdU immunoreactive neurons decreased in posterior horn and increased in anterior and lateral horn of superior thoracic segment of spinal cord in embronic rat.Given BrdU at E12d,most of BrdU immunoreactive neurons were mainly found in anterior and lateral horn of superior thoracic segment of spinal cord in embronic rat,on[1] [2] 下一页 上一个医学论文: Fos蛋白在LPS免疫激发大鼠脑内的分布 下一个医学论文: 镍和镉对人外周血淋巴细胞的DNA损伤作用
