类风湿关节炎患者红细胞C3b受体数目改变及基因多态性分析 |

【摘 要】 目的 分析类风湿关节炎(RA)患者C3b受体(C3bR)表达数目及C3bR 基因多态性分布规律，探讨C3bR数目改变的可能原因。方法 分别测定63例RA患者及健康献血员60人的红细胞C3b受体花环率(C3bRR)及免疫 复合物花环率(C3bICR)。用限制性片段长度多态(RFLP)分析及Southern blotting杂 交技术检测C3bR基因多态性。结果 RA患者红细胞C3b -RR和C3b-ICR分别显著低于和高于正常对照组。C3bR基因的两个等位基因基 因型和表现型频率在RA组和对照组均无显著差异。结论 RA患者C 3bR数目下降可能与基因变异无关，可能是受体基因转录和翻译过程异常所致。设法增 加红细胞C3bR数目以加速免疫复合物的清除，可能是治疗RA的有效途径之一。

【Abstract】 Objective To analyse the number o f erythrocyte C3breceptor (C3bR) and polymorphism of C3bR gene in patients with rheumatoid arthritis (RA) and to probe into the cause of chang es of C3bR number.Methods The erythrocytes C3bRR an d C3bICR in 63 patients with RA and 60 healthy volunteers were detected.Me anwhile,the polymorphism of C3bR gene were analysed with restriction fragm ent length polymorphisms (RFLP) and southern blotting technique.Results The numbers of C3bRR and C3bICR were significantly decreas ed and increased in RA patients than those in healthy volunteers respectively.Ho wever,there was no difference in the frequency of genotype and phenotype for the two alleles of C3bR between RA patients and healthy volunteers.Conclusion The lower number of erythrocyte C3bR in RA patients may not be caused by the C3bR gene polymorphism and could be caused by an abno rmality of transcription and translation for C3bR gene.The increase of the number of C3bR on erythrocyte to accelerate the clearance of immune complex may be a good way for the treatment of RA.
