ients with coronary heart disease and the control groups, either in genotypes or in alleles. The attributable risk of FABP2 Thr54(+) genotype for the type 2 diabetic patients with coronary heart disease was 25.6%, and odds ratio was 2.13 (95% confidence interval is 1.05~4.45). The patients of Thr54(+) genotype with coronary heart disease had significantly higher fasting insulin level and plasma triglyceride value than those of Thr54(-) genotype patients with coronary heart disease (P<0.01,P<0.05). The patients of Thr54(+) genotype had significantly lower insulin sensitive index and lower high density lipoprotein cholesterol (HDL-C) than those of Thr54(-) genotypeРin type 2 diabetic patients with coronary heart disease (P<0.05). Conclusions FABP2 gene polymorphism seems to be associated with the type 2 diabetic patients with coronary heart disease and their insulin resistance. 临床流行病学调查表明:糖尿病冠心病发病率是非糖尿病人群3~4倍。2型糖尿病合并冠心病患者存在明显的脂质代谢异常。 也有一些学者提出胰岛素抵抗是糖尿病冠心病脂质代谢异常的原发因素〔1〕。为进一步从分子水平探讨2型糖尿病合并冠心病的可能发病机制,我们就FABP2 54位密码子变异与2型糖尿病并冠心病的关系进行研究探讨。
对象和方法
一、对象 1.正常对照组(Control):60人(男29,女31),年龄(54±6)岁,无肝、肾、甲状腺疾病、无糖尿病和冠心病。 2.非糖尿病冠心病组(CHD):62例(男34,女28),年龄(53±8)岁,冠心病诊断按1979年WHO缺血性心脏病诊断标准。 3.2型糖尿病组:146例(男70,女76),年龄(58±10)岁,其中2型糖尿病无冠心病组(DM-1)64例,2型糖尿病并冠心病组(DM-2)82例。糖尿病诊断按1985年WHO诊断标准。 所有受检对象均是武汉地区汉族人,受检者之间无血缘关系。 二、方法 1.采用酚-氯仿法提取外周血基因组DNA。 2.PCR引物引自文献如下〔2〕:5 -ACAGGTGTTAATATAGTGAAAAG-3 ,3 -CTGCCCTTGAC-TTGAGTCCCAT-5 ,DNA扩增体系:在反应液中依次加入10×反应缓冲液(复华公司)5 μl,4种脱氧三磷酸核苷酸5 μl(各2 mmol/L)(华美公司),每种引物50 pmol/L,基因组DNA上一页 [1] [2] [3] 下一页 上一个医学论文: 高浓度胰岛素 葡萄糖对HepG2细胞蛋白激酶C活性的影响 下一个医学论文: 甲状腺激素对新生期大鼠下丘脑Go蛋白 亚单位的影响
