摘要:目的 观察干扰素γ(Interferon-γ,IFN-γ )对兔耳创面愈合期伤口周边组织和肉芽组织中蛋白酪氨酸激酶(Protein Tyrosine Kinas e, PTK)活性以及伤口愈合时间的影响,探讨IFN-γ外用于皮肤创面的信号传导机制。方法 在兔耳腹侧伤口外用IFN-γ,观察上皮化的时间并测定正常兔耳、 伤后1天、3天、6天和创面上皮化时(12~14天)组织中PTK活性变化。结果 周边和肉芽组织中PTK活性伤后均高于正常皮肤组织(P<0 .01),周边组织伤后6天即恢复正常水平,而肉芽组织在上皮化后仍然较高。IFN-γ可使伤 口周边和肉芽组织的PTK活性升高(P<0.05或0.01)。IFN-γ可使 伤口愈合延迟1.23天(P<0.01)。结论 创 伤愈合组织中PTK活性升高且与增生程度相关。PTK既介导生长刺激信号使细胞增殖和合成物 质的功能增强,也介导IFN-γ的信号使愈合延迟。 关键词:信号传导; 创伤愈合; 蛋白酪氨酸激酶; 干扰素-γ 中图分类号:R318.0 文献标识码:A 文章编号:1004-6526(2000)05-0266-03
Effects of interferon-γ on activity of protein tyrosine kin ase in wound margin tissue and granulation tissue of rabbit ear

Abstract:Objective By observing the changes of activity o f protein tyrosine kinase (PTK) in wound margin tissue and granulation tissue of rabbit ear and speed of wound healing after using IFN-γ to research the mechan ism of IFN-γ effecting. Methods IFN-γ was applied on the wound of rabbit ear. The PTK activity in the tissue from 1d, 3d, 6d after wound epithelization and in normal skin were measured by phosphorus (32p) incopo ration. The time of epithelialization was also observed. Results The PTK activity in wound margin tissue and granulation tissue was obviou sly elevated in comparing with that of normal skin (P<0.0 1). PTK activity of wound margin tissue recovered to basic level after 6d wh ile that of granulation tissue still maintained at higher level (P<0.05). IFN-γ could increase PTK activity(P<0.0 1) and delay wound healing(P<0.01). Conclusion PTK activity in wound healing might be related to healing rate.
