实验性消化道溃疡肠上皮内淋巴细胞体外转化能力与IFN 分泌活性的变化

  摘要 目的:了解消化道溃疡肠上皮内淋巴细胞(IEL)的转化及IFNγ的分泌能力。方法:利用水浸限制刺激诱发的小鼠消化道应激溃疡动物模型,采用〔3H〕TdR掺入法和ELISA法分别检测IEL的体外转化能力和IFNγ的分泌水平。结果:给予小鼠水浸限制刺激诱发消化道溃疡后3 d,IEL对有丝分裂原ConA刺激的体外转化能力最强,刺激指数为1.15,1周后降至正常。此时,IFNγ的分泌水平也最高,第6天尚未恢复到正常水平。结论:在应激性消化道溃疡发生后,IEL以其增强转化能力和细胞因子分泌水平对机体起到免疫保护作用。

The Changes of Proliferation and Interferon-γ Secretion of Intestinal Intraepithelial Lymphocyte from Mice with Experimental Digestive Ulcer

  ABSTRACT Objective:In order to understand the ability of the proliferation and INFγ secretion of intestinal intraepithelial lymphocytes (IELs) in digestive ulcer.Methods:[3H]TdR incorperation and ELISA techniques were used to examine the ability of proliferation and INFγ secreting level of IEL in the animal model with experimental digestive ulcer induced by water immersion restrain stress. Results:The proliferation of the IELs to the stimulation of mitogen ConA was significantly enhanced and the stimulation index was increased to 1.15 on the third day after the stress, and restored to the normal level after a week; The level of IFNγ secreted by the IELs was also the highest on the same day and could not restore completely to the normal even on the sixth day after the stress. Conclusion:The IELs were activated in digestive ulcer and considered to be important in the intestinal mucosa immunity.

  肠上皮内淋巴细胞(intestinal intraepithelial lymphocyte, IEL)是存在于消化道肠上皮细胞间的特殊T淋巴细胞。IEL数量庞大,小鼠小肠内IEL数量约5×107~7×107个,相当于末梢淋巴组织中T细胞总数的一半[1]。与末梢淋巴组织中的CD8αβ+T细胞不同,IEL多为表达CE8αα+的T细胞;50%的IEL,T细胞表面抗原为TCRγ/δ+,白细胞表面抗原为CD-5[2,3]。I
