子宫内膜异位症患者外周血单核细胞的细胞因子测定 |

【摘要】 目的 探讨子宫内膜异位症(内异症)患者外周血单核细胞分泌的白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、肿瘤坏死因子α(TNF-α)和一氧化氮( NO)水平的变化及其意义。方法 从31例内异症患者（轻度组11例，为Ⅰ、Ⅱ期；重度组20例，为Ⅲ、Ⅳ期)和15例对照者(对照组，为行输卵管结扎术和吻合术患者)的外周血中分离出单核细胞，体外经脂多糖、植物血凝素诱生，培养48小时收集上清液，用酶联免疫吸附法测定IL-6、IL-8和TNF-α的水平，用镀铜镉还原法测定反映NO水平的亚硝酸基/硝酸基(NO-2/NO-3)水平。结果 轻度组和重度组IL-6、IL-8、TNF-α及NO水平明显高于对照组，差异有极显著性(P＜0.01)；轻度组和重度组之间差异无显著性(P＞0.05)；内异症患者IL-6与IL-8、TNF-α与NO水平呈明显正相关(P＜0.01)。结论 内异症患者外周血单核细胞免疫功能紊乱，处于激活状态并产生过量的IL-6、IL- 8、TNF-α及NO，可能共同参与内异症的发病。

Study on in Vitro Cytokines Levels Induced from Peripheral Mononuclear Cells in Patients with Endometriosis

【Abstract】 Objective To study the changes and significance of interleukin-6 ( IL- 6)，interleukin-8(IL-8)，tumor necrosis factor-α ( TNF-α)and nitric oxide (NO) levels in vitro cultured system of peripheral mononuclear cells in patient with endometriosis(Em). Methods Peripheral blood mononuclear cells from 31 cases ofendometriosis, classified into mild group (stage Ⅰ and Ⅱ, n＝11), and severe group (stage Ⅲ and Ⅳ, n＝20) according to the Revised American Fertility society criteria 1985, and 15 normal controls were separated and induced by lipopolysaccharide and phytohemagglutinin, followed by 48 hour incubation in vitro, then the supernatant were collected. The levels of IL-6, IL-8 and TNF-α were measured by enzyme linked immunosorbent assays, the concentrations of NO, expressed by nitrite and nitrate (NO-2/NO-3) content were measured by Griess methods. Results The concentrations of IL-6, IL-8, TNF-α and NO were significantly higher in both mild and severe Em groups than those in controls (P<0.01).
