糖皮质激素对大鼠未成熟肺PDGF mRNA表达的调节 |

【摘要】 目的 进一步提高糖皮质激素(GC)预防新生儿呼吸窘迫综合征(NRDS)的效果。方法 用溶液杂交技术观察血小板源生长因子(PDGF)及其受体(PDGFR)mRNA在大鼠不同成熟阶段肺中的表达,以及GC对二者在肺内表达的调节。结果 (1)PDGF-BB、PDGFR-α mRNA随胎龄增长而增高,高峰在19~21天,新生时表达减少,成年时又增高到接近胚胎晚期水平;PDGF-AA和PDGFR-β mRNA在胚胎晚期、新生与成年鼠肺中均有表达,不随胎龄增长而变化,肺内PDGFR-β的表达高于PDGFR-α。(2)GC可上调18天胎肺中PDGF-BB、PDGFR-α mRNA的表达,而下调19、21天胎肺中它们的表达。GC对胎鼠肺组织PDGF-AA、PDGFR-β mRNA的表达无明显影响。结论 PDGF与PDGFR mRNA随胎龄增长而变化,与胎肺发育密切相关;GC对大鼠未成熟肺PDGF-BB及PDGFR-α的基因表达有调节作用,在胎肺发育过程中激素与生长因子间有某种联系,以维持胚胎肺的正常发育。 【关键词】 糖皮质激素类 血小板源生长因子 RNA,信使 基因表达 大鼠 肺
The effects of glucocorticoid on platelet-derived growth factor mRNA expression in fetal rat lungs CHEN Ling, YANG Yi, JIN Qinli, et al. Institute of Pediatrics, Children′s Hospital, Shanghai Medical University, Shanghai 200032 【Abstract】 Objective To study (1) differential expressions of platelet-derived growth factor (PDGF) and platelet-derived growth factor receptor (PDGFR) mRNA and (2) regulation of dexamethasone (Dex) on the expression of these growth factors in late lung development of fetal rats. Methods RNase protection assays were used to study mRNA expressions of PDGF-AA PDGF-BB chains and their cognate receptors during late lung development in fetal rats. The mRNA was extracted from lungs of 18,19 and 21 days of gestation (term=22 days), newborn and adult rats. By serum free cultures of fetal lung explants, the effects of Dex on PDGF′s expression were also investigated. Results Expressions of PDGF-BB chain and PDGFR-α mRNA were low at the pseudoglandular stage of day 18, reached the peak level at the canalicular stage of day 19 and the sacculate stage of day 21 of gestations, fell again in newborn rats, and then increased in adult rats. Expressions of PDGF-AA chain and PDGFR-β mRNA did not change significantly with the advancing gestation.
