异丙酚对过氧化氢所致的血管内皮细胞损伤的保护作用 |

摘要 目的:观察异丙酚对过氧化氢(H2O2)介导的内皮细胞损伤的作用。方法:按异丙酚的不同浓度,以脂肪乳剂为对照,用2mmol/L H2O2导致内皮细胞损伤,将实验分为10组。检测乳酸脱氢酶释放率(LDH)及硫代巴比妥酸反应产物(TBARS)含量,超氧化物岐化酶(SOD)和过氧化氢酶(CAT)活性。并观察细胞形态学改变。结果:临床相关浓度的异丙酚明显降低H2O2所致的LDH释放,抑制TBARS的产生,保护SOD和CAT活性(P<0.001)。减轻了细胞形态及超微结构的改变。结论:异丙酚对H2O2损伤的内皮细胞具有保护作用。 关键词 异丙酚 内皮,血管的 细胞 过氧化氢 创伤和损伤 保护
Protective effects of propofol on hydrogen peroxide-induced injury in bovine aortic endothelial cells Wang Yaping,Chang Yetian,Zhong Lin. Department of Anesthesiology,The Second Affiliated Hospital,Hunan Medical University,Changsha 410011 Abstract Objective:To observe the effects of propofol on the endothelial cells injured by hydrogen peroxide.Method:Endothelial cells injured by 2mmol/L hydrogen peroxide were divided into ten experimental groups according to the different concentrations of propofol,using intralipid as control.The release of lactate dehydrogenase (LDH),production of thiobarbituric acid reactive substances (TBARS),activities of superoxide dismatase(SOD) and catalase (CAT) were determined as well as morphological change.Result:Propofol at clinical relevant concentrations not only decreased the release of LDH and production of TBARS,but also protected significantly the activities of SOD and CAT (P<0.0001).Conclusion:Propofol can protect vascular endothelial cells against oxidant damage. 异丙酚是一种静脉麻醉药,近年来,其抗氧化作用已逐渐被人们认识,但异丙酚减轻氧化损伤作用的强弱有分岐,对血管内皮细胞氧化损伤的作用还未见报道。本实验观察异丙酚对过氧化氢介导的内皮细胞损伤的作用,为临床麻醉提供理论依据。
材料与方法
一、主要药品及试剂:过氧化氢(分析纯)(上海桃浦化工厂);异丙酚(英国捷利康医药公司);脂肪乳剂(华瑞制药有限公司);DMEM培基和胰蛋白酶(Gibco,USA);胶原酶Ⅱ型(Sigma,USA)。 二、牛主动脉血管内皮细胞培养:按Ryan的方法进行原代培养并传代〔1〕。取第五代细胞作透射电镜观察发现WebelPalade小体,鉴定为血管内皮细胞〔2〕,取5~10代细胞实验。 三、生化及酶学指标的测定 1.实验分组:将细胞接种于24孔培养板,48小时后细胞密度为1×105个/孔,分为10组,①正常对照组;②过氧化氢组(H2O2组);③脂肪乳剂组(IN组);④脂肪乳剂和过氧化氢组(H2O2+IN组);⑤12.5μmol/L异丙酚组(P12.5组);⑥12.5&[1] [2] 下一页 上一个医学论文: 前列腺偶发癌的发病及治疗探讨 下一个医学论文: 创伤性休克及复苏后肠粘膜免疫功能的改变
