未成熟树突细胞诱导同种免疫低应答性的研究

  摘 要 目的:证实缺少辅助刺激分子的未成熟树突细胞(DC)在体外诱 导同种T细胞的低应答性。方法:应用骨髓微血管内皮细胞作为饲养细胞扩增同品系小鼠骨髓细胞 ,经不同浓度GM-CSF作用,在体外培养出不同发育阶段的DC。采用初次和再次单向混合淋 巴细胞培养方法观察Balb/c小鼠脾脏T细胞对同种未成熟DC或脾细胞的应答能力。结果:在初次MLR试验中,Balb/c T细胞对C57BL/6小鼠未成熟DC刺激只 呈现出微弱的增殖反应,SI=2.84±0.60,明显低于对相应品系鼠成熟DC或脾细胞的增殖 反应(SI分别为15.72±6.49,6.57±2.85);在再次MLR系统中,经未成熟DC预处理的Ba lb /c T细胞对C57BL/6来源的脾脏细胞只发生低应答反应(SI=1.29±0.19),而对无关品系AK R的脾脏细胞仍显示较高的增殖能力(SI=10.64±1.89)。结论:缺乏辅助刺激分子的未成熟DC在体外MLR系统中诱导出同种Balb/ c小鼠T细胞免疫低应答性。

Experimental study on induction of allo-hypo-immuno- responsiveness by immature dendritic cells

Abstract Objective:To testify the induction of allo -hypo-immuno-responsiveness by immature dendritic cells(DC)with deficiency of costi mulators.Methods:Bone marrow microvessel endothelial cell s (BMEC)from C57BL/6 mice were used as feed cells.Bone marrow cells from the sam e strain were expanded in this culture without differentiation.Those expanded bo n e marrow cells would differentiate to dendritic cells at different stages accord ing to the concentrations of GM-CSF.The primary and secondary mixed lymphocyte reaction(MLR)assay was used to be a model in vitro to analyze response of Balb/c T cells to allogenic immature DC or splenocytes.Results:Balb/c T cells proliferated weakly to C57BL/6 imma ture DC in primary MLR,SI=2.84±0.60,which was extremely lower than to C57BL/6 mature DC or splenocyte(SI=15.72±6.49,6.57±2.85 respectively);in secon dary MLR,prime d by immature DC,Balb/c T cells showed low responsiveness to C57BL/6 splenocyte( SI=1.29±0.19),while a strong proliferation to AKR could be detected(SI=10.64 ±1.89).Conclusion:Immature DCs with deficiency of costi m ulators could induce an allo-hypo-immuno-responsiveness of Balb/c mic
