【摘要】 目的 探讨注射用紫杉醇脂质体对人子宫内膜癌细胞系的体外杀伤作用。方法 以不同浓度含紫杉醇脂质体的培养基作用于人子宫内膜癌细胞系Ishikawa细胞，并设对照(普通培养基)，通过MTT、倒置显微镜观察细胞增殖状况及形态，流式细胞分析技术检测该药对细胞生长的影响。结果 经不同浓度紫杉醇脂质体作用24～96h，细胞存活率显著下降(P<0.05);镜下观察显示部分细胞皱缩变形，崩解坏死，细胞脱壁悬浮;流式细胞术结果显示细胞凋亡率逐渐增加，细胞周期各时相中，S期比例减少。结论 紫杉醇脂质体可抑制子宫内膜癌Ishikawa细胞的生长。

Methods The human endometrial carcinoma Ishikawa cell line was cultured with different concentrations of the liquid form of liposome encapsulated paclitaxel. MTT assay, inverted microscopic observation and flow cytometry assay (FCM) were then used to monitor the growth and proliferation of the cell line. Results When treated with different concentrations of liposome encapsulated paclitaxel for 24 to 96 hours, Ishikawa cells survival rate decreased significantly (P<0.05); degeneration and necrosis of Ishikawa cells were found under microscope. The rate of cell apoptosis was increased and the rate of S phase was reduced. Conclusion Liposome encapsulated paclitaxel can inhibit endometrial carcinoma cells.

紫杉醇(paclitaxel)是一种存在于紫杉烷类植物树皮和枝叶中的二萜烯类物质，能够稳定细胞微管阻止细胞分裂而抑制癌细胞生长。临床研究表明，紫杉醇对多种实体肿瘤疗效显著。但其水溶性差，聚氧乙基代蓖麻油和无水乙醇组成的混合溶媒会引起组胺的释放，易导致严重的过敏反应[1]。脂质体作为一种新型的药物载体，具有靶向性和缓释性，可提高疗效，降低毒性[2]，是目前研究的热点。本研究探讨紫杉醇脂质体对子宫内膜癌Ishikawa细胞系的体外杀伤作用，为临床应用提供实验依据。
1 材料与方法
1.1 癌细胞株 Ishikawa细胞株系北京大学人民医院妇产科实验室保存并传代培养。
