含何首乌血清对人胚肺二倍体成纤维细胞端粒酶活性的影响

【摘要】 目的 观察含何首乌血清对人胚肺二倍体成纤维细胞(2BS)超氧化物歧化酶(SOD)活性、c-myc mRNA表达及端粒活性的影响。方法 采用含何首乌血清对2BS细胞株进行处理,用黄嘌呤氧化酶法检测SOD活性,RT-PCR检测c-myc mRNA表达,TRAP-ELISA检测端粒酶活性。结果 与正常对照组相比,用药组2BS细胞SOD活性明显增加(P<0.05),c-myc mRNA表达无变化,端粒酶活性显著增加(P均<0.05)。结论 含何首乌血清对2BS细胞具有抗衰老作用,其机理可能与促进自由基的清除和激活端粒酶活性有关。

  Effects of Tuber Fleeceflower Root serum on telomerase activity of human fetal lung diploid fibroblasts

  Abstract: Objective To observe effects of Tuber Fleeceflower Root serum on activity of SOD, the expression of c-myc mRNA and the activity of telomerase of human fetal lung diploid fibroblasts (2BS). Methods 2BS cell lines were treated with Tuber Fleeceflower Root serum. The activity of c-myc mRNA was detected by semi-quantitive RT-PCR. A TRAP-ELISA method was used to measure the telomerase activity. Results Compared with those in the control group, the activity of SOD increased obviously (P<0.05), the expression of c-myc mRNA remained on the same level, and the telomerae activity were enhanced significantly(P<0.05) in the drug group. Conclusion Tuber Fleeceflower Root serum can postpone senescence of 2BS, its mechanism may be associated with enhancement elimination of free radicals and activation of telomerase.

  何首乌又称首乌、赤首乌、乌肝石等,为蓼科植物何首乌(Polygonum multiflorum Thunb.)的块根,具有补肝肾、益筋血、壮筋骨、乌须发之功效。研究表明[1],何首乌具有抗衰老、保护神经、增强免疫功能、降低血脂、抗动脉粥样硬化、抗心肌缺血、保肝、抗癌、抗菌等作用。在正常体细胞中,由于端粒的缩短及缺乏端粒酶的表达,细胞逐渐衰老。仅有极少数的细胞因为端粒酶在某种因子的作用下偶然激活,持续合成端粒DNA序列使细胞绕过M2退化期,获得无限增殖能力,从而逃避了程序性老化。端粒酶活性的调节可为抗衰老的研究提供一条新途径[2]。本组研究何首乌血清药物对人胚肺二倍成纤维细胞(2BS)内超氧化物歧化酶(SOD)活性、c-myc表达及端粒酶活性的影响,以探讨其抗衰老的可能机理,为抗衰
