蝌蚪提取液诱导HL60细胞分化和凋亡的实验研究

  【摘要】 目的 为了寻找新的肿瘤细胞分化诱导剂,探讨蝌蚪醚提取液(T8712)的抗肿瘤作用。 方法 将HL60细胞培养于T8712和RPMI1640按1:200(v/v)混合的培养基中。 结果 T8712能诱导HL60细胞向单核/巨噬细胞方向分化,表现为生长抑制,NBT还原能力提高,酸性非特异性酯酶(ANAE)活性增加,形态学观察类似单核/巨噬细胞。
  当T8712与RPMI1640培养基按1:100(v/v)混合则可诱导HL60细胞凋亡,如出现凋亡的形态学特征(凋亡小体出现等);DNA琼脂糖凝胶电泳显示典型的阶梯型带;流式细胞式分析证明凋亡细胞数增加,但Bcl-2蛋白未有明显改变。 结论 T8712能抑制HL60细胞增殖,在不同浓度条件下可诱导HL60细胞向单核/巨噬样细胞分化或发生凋亡,其凋亡过程可能与Bcl-2无明显相关性。

THE INDUCTION OF DIFFERENTIATION AND APOPTOSIS IN
HL60 CELLS BY TADPOLE EXTRACT

  【Abstract】 Objective In order to search for new tumor cell differentiation inducer, the antitumor effects of the tadpole ether extract (T8712) were studied. Methods T8712 and RPMI1640 medium were mixed in the ratio of 1:200(v/v). The HL60 cells were cultured in this mixed medium. Results T8712 might induce HL60 cell differentiation to monocyte-macrophage, as shown by growth inhibition; enhancing of NBT reducing ability; increasing of ANAE activity, similar shape to monocyte-macrophage.
When the ratio of T8712 and RPMI1640 medium is raised to 1:100(v/v), HL60 cells were induced to apoptosis, as evidenced by (1) characteristic morphology of apoptosis (i.e, formation of apoptotic bodies, etc). (2) DNA agarose gel electrophoresis showed a characteristic “ladder” pattern. (3) The flow cytometry analysis showed the number of apoptotic HL60 cells increased, but Bcl-2 proteins did not change. Conclusion T8712 might inhibite HL60 cell proliferation. Under distinct circumstances T8712 might induce HL60 cell differentiation to monocyte-macrophage or apoptosis, but the apoptotive course was not striki
