海马内微量注射硫酸镁对马桑内酯诱发大鼠痫样放电的影响

  摘 要 目的和方法:采用皮层应用或海马内注射马桑内酯的方法致痫大鼠,用钙离子选择性微电极检测了致痫动物皮层、海马细胞外Ca2+浓度的变化,观察了海马CA2区注射硫酸镁对致痫大鼠脑电图和皮层、海马诱发电位的影响。结果:马桑内酯致痫时,皮层、海马细胞外Ca2+浓度分别降低了0.61 mmol.L-1和0.74 mmol.L-1,海马内注射硫酸镁能明显地抑制致痫动物皮层、海马诱发电位和脑电图痫样放电。结论: 镁盐的上述作用可能与抑制Ca2+流入神经元内有关。

Effect of microinjection of magnesium sulphate into hippocampus
on epileptiform discharges induced by coriaria lactone in rats

  Abstract  AIM and METHODS:In the experiments, application of coriaria lactone(CL) to cerebral cortex caused the epileptiform discharges in rats.The calcium ion selective microelectrode was used to measure changes of extracellular calcium ion concentration([Ca2+]o) of cerebral cortex and hippocampus during epileptic discharges induced by CL.The electrocoticogram(ECoG)and the evoked potentials of cerebal cortex and hippocampus were observed after the microinjection of magnesium sulphate into hippocampal CA2 area in epileptic rats. RESULTS: The [Ca2+]o of cerebral cortex decreased by 0.61 mmol.L-1 and that of hippocampus by 0. 74 mmol.L-1(P<0.01) in epileptiform rats. The intrahippocampal injection of magnesium sulphate markedly inhibited the evoked potentials of cerebral cortex and hippocampus and the epileptiform discharges from ECoG. CONCLUSION: The inhibitory effect of magnesium sulphate might be related to restraining the Ca2+ influx into neurons.

  致痫动物大脑皮层或海马脑片神经元发生异常放电可被有机钙通道阻滞剂verapamil、尼莫地平等所抑制[1,2,4],表明癫痫的病理过程与神经元跨膜钙内流有关。我们采用皮层贴敷浸有马桑内酯(coriaria lactone,CL)滤纸或海马内注射CL的方法造成大鼠皮层及海马神经元异常放电的癫痫模型,用钙离子敏感性微电极(Ca2+-ISE)检测皮层和海马细胞外钙离子浓度的变化,并以平均诱发电位和皮层脑电图(ECoG)为指标,观察海马内注射
