关于p38及p27在大肠癌中的表达及其临床意义 |

【摘要】 目 的:探讨人大肠癌中有丝分裂原激活蛋白激酶p38和细胞周期负性调控因子p27表达的临床意义。方法:应用SABC法检测46例人大肠癌组织及其正常大肠组织(距肿瘤>5 cm)中p38蛋白和p27蛋白的表达情况。结果:p38在大肠癌中的阳性表达率为95.65%，在正常大肠组织中阳性表达率为39.13%,定位于胞浆，胞核无表达,两者差异有显著性(P<0.01); p27在大肠癌中的阳性表达率为56.5% ,在正常大肠组织中阳性表达率为86.96%,主要定位于胞核,仅少量胞质表达, 两者差异有显著性(P<0.01)。p38和p27的表达都与患者年龄、性别、肿瘤大小、肿瘤部位没有相关性(P>0.05),与Dukes分级、肿瘤分化程度及淋巴结转移有密切相关，差异有显著性(P<0.01)。结论: 大肠癌组织中p38蛋白过表达及p27蛋白低表达都与大肠癌的发生、发展和转移密切相关，推测其可能作为反映恶性肿瘤生物学行为及预测预后的指标;p38可能是促进p27降解的一种负性调节因子。

Abstract:Objective To investigate the clinical significance of p38MAPK expression and cell-cycle-negative-regulator p27 expression in human colorectal carcinomas.Methods The samples were obtained from 46 cases . p38MAPK protein and p27 protein expression was detected by immunohistochemistry using the SABC staining method at the two sites: carcinoma tissue and its peripheral nomal tissue (5 cm away from tumor).Results Expression of p38MAPK protein was mainly localized in the cell cytoplasm and never in the nucleus and expression of p27 protein was mainly localized in the cell nucleus and only rarely in the cytoplasm . Positive rate of p38MAPK protein expression was 95.65% in carcinoma tissues,33.13% in its nomal tissue;Positive rate of p27 protein expression was 56.5% in carcinoma tissues,87.0% in its nomal tissue.p38MAPK and p27 protein expression in carcinoma tissue was correlated significantly with dukes stage (P<0.01),histology grade (P<0.01) and local lymph node involvement (P<0.01) ,but not with age ,gender,site or size of tumor (P>0.05).Conclusion p38 and p27 may play an important role in the occurrence，development an [1] [2] [3] [4] [5] 下一页
