【摘要】目的 研究骨髓基质细胞(BMSCs)移植对大鼠脊髓损伤(SCI)后神经细胞凋亡的影响,探讨骨髓基质细胞移植治疗脊髓损伤的机制。方法 取大鼠股骨和胫骨骨髓培养BMSCs并传代。参照Taokas方法制作大鼠脊髓压迫损伤模型。脊髓损伤后30 min,进行BMSCs或DMEM培养液损伤周围局部四点注射。在术后3、7和14 d,应用TUNEL方法检测脊髓损伤周围神经细胞凋亡情况。结果 脊髓损伤后,损伤区周围组织内TUNEL阳性细胞数量明显增加,阳性细胞分布于脊髓灰质和白质内,但以白质内神经细胞为主。与DMEM组大鼠相比,移植术后3、7和14 d,BMSCs移植显著减少脊髓损伤周围区凋亡的神经细胞数目(P<0.05)。结论 脊髓损伤后,BMSCs移植能够抑制神经细胞凋亡。

Effects of Bone Marrow Stromal Cells Transplantations on Apoptosis of Neural Cells after Spinal Cord Injury in Rats

Abstract:Objective To study the effects of Bone Marrow Stroma Cells(BMSCs) transplantation on apoptosis of neural cells after spinal cord injury(SCI),and to approach on the mechanism of repairing the SCI by BMSCs transplantation.Methods Obtained from rat femurs and tibias, BMSCs are cultivated and generated. The SCI model was made by consulting the Taoka s method. Rats were received four injections at peri-lesion area of BMSCs or DMEM culture fluid at 30 minutes after SCI. The change of apotosis was observed with TUNEL method in spinal cord at 3, 7 and 14 days after the transplantation. Results The numbers of TUNEL-positive cells increased approximately 1 to 2 mm from the injury epicene after SCI. The TUNEL-positive cells were present in both gray and white matter of the Spinal cord, but mainly in white matter. With BMSCs transplantation, the number of TUNEL-positive cells around the lesion epicenter were significantly reduced compared with that of DMEM treatment at 3, 7 and 14 day after transplantation (P<0.05). Conclusions BMSCs transplantation may inhibit apoptosis of neural cells in spinal cord tissue after spinal cord injury (SCI).
