声敏剂血卟啉对COC1 DDP细胞顺铂敏感性的影响及机制探讨

【摘要】 目的 探讨声敏剂血卟啉对COC1/DDP细胞顺铂敏感性的影响及其机制。方法 将人卵巢癌顺铂耐药细胞株COC1/DDP分为A、B、C、D 4组(分别为对照组、单纯顺铂组、血卟啉+超声组、顺铂+血卟啉+超声组),观察各组处理后癌细胞的增殖、细胞克隆形成情况,联合用药效果及bcl2和bax表达的变化。结果 D组COC1/DDP细胞增殖抑制明显,细胞凋亡率升高,均与其他组差异有统计学意义(P<0.05),bcl2蛋白阳性表达率在C、D组显著降低(P<0.05),A、B组差异无统计学意义(P>0.05)。bax蛋白阳性表达率在各组表达差异无统计学意义(P>0.05)。结论 声动力疗法联合顺铂能有效地增强对耐药卵巢癌细胞的杀伤能力,减少癌细胞的生成。

  A Discussion on Effect and Mechanism of Haematoporphyrin to Chemotherapy Sensitivity of Cisplatin on COC1/DDP

  【Abstract】 Objective To determine whether sonodynamic treatment (SDT) can enhance chemotherapy effect of cisplatin on ovarian cancer cells. Methods Drug resistance ovarian cancer cells COC1/DDP were divided into 4 groups randomly:cisplatin (DDP)group,haematoporphyrin(Hp)+ Ultrasound group,cisplatin(DDP)+ haematoporphyrin(Hp)+Ultrasound group and control group.Cell proliferation,apoptosis,protein bcl2 and bax expression were observed.Results In DDP+ Hp+Ultrasound group,cell proliferation was greatly inhibited,COC1/DDP cell clone efficiency was the lowest(P<0.05).Bcl2 expression in Hp+ Ultrasound group and DDP+Hp+ Ultrasound group deceased significantly (P<0.05),while there were no obvious difference in control goup and cisplatin group.We did not observe great difference in protein bax expression in any group (P>0.05). Conlusion SDT could effectively enhance cell killing ability of cisplatin on drug resistance ovarian cancaer cells.
