肾综合征出血热病人恢复期血清对汉滩病毒感染体外培养神经元的保护作用 |

【 摘要 】 目的 用体外培养的昆明小鼠胚胎脑皮质神经元，感染汉滩病毒后，加入肾综合征出血热(HFRS)病人恢复期血清，观察病毒感染神经元后细胞的变化以及HFRS病人恢复期血清对神经元的保护作用。 方法 将病毒感染的神经元加入不同浓度的HFRS病人恢复期血清(同时设立阴性血清对照组及正常细胞对照组和感染病毒对照组)，使用MTT比色试验检测神经元存活情况。 结果 各组神经元活性的MTT比色试验显示，病毒感染组与未感染对照组比较差异有非常显著性(P<0.001)，阴性血清对照组与正常细胞对照组比较，差异也有统计学意义(P<0.01)。HFRS病人恢复期血清保护组与阴性血清对照组各小组间比较差异也都有非常显著性(P<0.001)。 结论 HFRS病人恢复期血清具有一定的中和病毒保护培养的神经元作用。

【 Abstract 】 Objective Convalescent serum from patient of hemorrhagic fever with renal syndrome (CSPH) was added to the HTNV infected embryo mouse brain neurons to test the protective action of CSPH on HTNV. Methods Morphological changes were observed with Phase-contrast microscopy and the viability of neuron cells were examined with MTT colorimetric assay. Results Significant difference of the A value formd between the HTNV infected neuron cell cultures and uninfected normal control cultures (P＜0.001). While no obvious differences were observed among different serum dose groups both in the convalescent serum protection group and in the negative serum control group (P＞0.05), significant differences of the A value were formed between the convalescent serum groups and the negative serum groups (P＜0.001). Conclusions Convalescent serum from patient with HFRS may produce protective action to the mouse brain neuron culture cells infected with HTNV.

肾综合征出血热(hemorrhagic fever with renal syndrome.HFRS)患者可出现神经系统症状，动物实验中乳鼠常常表现出严重的脑损伤〔1〕。以往对HFRS脑损伤的报道仅限于神经细胞的变性坏死和炎症细胞浸润等。缺少有关HFRS恢复期病人血清对病毒感染的神经元保护作用方面的报道。本研究利用体外培养的小鼠胚胎脑皮质神经元感染汉滩病毒(Hantaan virus,HTNV)后，加入HFRS病人恢复期血清，观察其对HTNV感染神经元的保护性作用。
