摘 要 目的:在裸鼠活体水平上观察bcl-2反义寡聚核苷酸(antisense oligodeoxynucleotide, ASON)对鼻咽癌细胞株CNE2-baxα凋亡的影响。方法:DNA合成仪合成22nt ASON片段,RT-PCR检测bcl-2表达,电镜观察细胞凋亡。结果:反义bcl-2 DNA片段局部注射CNE2-baxα致瘤组织,RT-PCR检测肿瘤组织中bcl-2表达被封闭,电镜观察视野下可见大量癌细胞凋亡,癌细胞数目减少,细胞间组织大量纤维化;凋亡的癌细胞电子密度增加,具典型凋亡特征。反义bcl-2封闭1周,抑瘤率74.4%,第2周为46.0%。结论:反义bcl-2抑制鼻咽癌细胞株CNE2-baxα致瘤性,促进细胞凋亡。 主题词 基因;鼻咽肿瘤;寡核苷酸类,反义;小鼠,裸
Antisense bcl-2 DNA enhances apoptosis of nasopharyngeal carcinoma cell line CNE2-baxα habouring in nude mice

Abstract AIM: To observe the effect of bcl-2 antisense oligod eoxynucleotide (ASON) on nude mice habouring tumor by inoculated human nasopharyngeal carcinoma cell line CNE2-baxα. METHOD: 22nt bcl-2 ASON was synthesized using DNA synthesizer. Apoptosis of neoplastic cells were observed with electron microscopy. RESULTS: Tumors were growing after CNE2 baxα cells were subcutaneously inoculated in nude mice. Suppression of bcl-2 expression was verified with RT-PCR assay after bcl-2 ASON was injected into the tumors. We also identified by electron microscopy that after ASON treatment large amount of tumor cells underwent apoptosis and the number of tumor cells reduced. The electron density of apoptotic cells was increased and apoptotic bodies formed.7 days after antisense bcl-2 DNA injection, the tumor-inhibiting rate was 74.4%, 14 days after, the rate was 46.0%. CONCLUSION:Antisense bcl-2 DNA inhibits the CNE2-baxα tumor growth and enhances apoptosis of CNE2-baxα cells. MeSH Genes; Nasopharyngeal neoplasms; Oligonucleotides, antisense; Mice, nude
