SD乳鼠窦房结细胞原代分散培养的光电镜研究

  【摘要】 目的 探索体外窦房结细胞原代纯化培养方法,观察培养的窦房结细胞生长状况及光电镜形态学特征。 方法 (1)用差速贴壁和Brdu处理作细胞培养;(2)观察和记录各类型培养细胞的生长状况;(3)HE染色和Heidenhain苏木素染色;(4)透射电镜观察。 结果 在培养的窦房结细胞中,至少有3种自发性收缩细胞及少量的星状和不规则细胞。收缩性细胞中,最多的是多边形细胞,其次是梭形,最少的是三角形细胞,且多边形细胞收缩频率最低(75.27±1.73/min ±s),细胞染色和电镜观察结果显示:多边形细胞的细胞器相似于原位窦房结组织中的起搏(P)细胞,梭形细胞相似于移行(T)细胞,P细胞核旁较易见到电子致密颗粒和中心粒,也常见细胞核染色体正处于细胞周期的分裂期的前、中期。 结论 多边形细胞很幼稚,具有不断分裂增埴的能力。

SINOATRIAL NODE CELLS IN PRIMARY DISSOCIATED
CULTURE FROM THE NEONATAL SD RATS

【Abstract】 Objective The purpose of the study was to develop primary pure-culture of the sinoatrial node(SAN) cells in vitro and to investigate the growth conditions,morphological criteria of light and eletron microscopy of cultured cells. Methods (1)Cell culture with differential attachment technique and BrdU-treated.(2)Observing and recording the growth condition of cultured cells of each type.(3)HE staining and Heidenhain hematoxylin staining for facilitating morphologic identification of cultured nodal cells.(4)TEM observation for demonstrating of cultured cells of SAN.Results Among the cultured cells of SAN,at least three distinctly different types of spontaneously contracting cells were observed,as well as a few stellar and irregular cells.The greatest proportion of contracting cells were relatively large polygonal cells and the second group of cells was those with spindle configuration and the last group of cells was triangle cell.The mean frequency(75.27±1.73/min ±s)of spontaneous contraction of the polygonal cells was lowest in all contraction cells.The results of cells staining and ultrastructure observation in cultured cells indicated that the characteristic of polygonal and spindle cells were separately similar to these i
