N 乙酰半胱氨酸和川芎嗪对博莱霉素诱导的肺纤维化和TGF 的影响

[摘要] 目的:探讨抗氧化剂N-乙酰半胱氨酸(NAC)和川芎嗪对博莱霉素所致小鼠肺间质纤维化的治疗作用及机制。方法:博莱霉素(BLM)制成小鼠肺间质纤维化模型。各组动物处死后提取肺组织和支气管肺泡灌洗液(BALF),进行病理半定量分析,检测谷胱甘肽(GSH)和丙二醛(MDA)含量、BALF中转化生长因子-β(TGF-β)水平。结果: NAC和川芎嗪治疗组肺泡炎、纤维化病变均较模型组减轻,7 d时肺组织GSH量较模型组高(P<0.05), MDA含量较模型组低(P<0.05),7、14 d时TGF-β蛋白水平较模型组低(P<0.05)。结论:NAC和川芎嗪可提高肺组织GSH含量,降低MDA生成,使肺泡炎和肺纤维化病变减轻。NAC和川芎嗪通过抗氧化作用,减少TGF-β产生,是其治疗肺纤维化的机制之一。

  The Effect of N-acetylcysteine and Tetramethylpyrazine on Bleomycin-induced Pulmonary Fibrosis and TGF-β

  Abstact: Objective To investigate the therapeutic effect of N-acetylcysteine and tetramethylpyrazine in bleomycin-induced pulmonary fibrosis in mice and explore its mechanism.Methods Fifty male Kunming mice were randomly categorized into four groups:the bleomycin(BLM) group,the N-acetylcysteine(NAC) treated group, the Tetramethylpyrazine treated group and the control group.Pulmonary fibrosis model was induced by intratracheal instillation of single dose BLM 5 mg?kg-1 in mice. NAC and Tetramethylpyrazine treated group were given with NAC 80 mg?kg-1 and Tetramethylpyrazine 40 mg?kg-1, respectively.The animals of the above three groups were killed on the 7th,the 14th and the 28th day after intervention, respectively. The mice in control group was instillated with saline intratracheally and killed on the 7th day.Pathological changes,levels of glutathione (GSH) and Malonyldialdehyde(MDA) in lung tissue,transforming growth factor-β(TGF-β) protein levels in BALF were determined. Results Attenuation of alveolitis and lung fibrosis after treatment with NAC and Tetramethylpyrazine was shown in pathogocal section(P<0.05). GSH was increased and MDA was decreased in lung tissue on the 7th day(P<0.05) in NAC and Tetramethylpyrazine treated group. TGF-β in BALF
