摘要:目的 制备PLGA-bBMP,PLGA-bBMP-IGF-Ⅱ,PLGA-bBMP-IGF-Ⅱ-bFGF三种诱导骨再生膜并比较其修复兔桡骨骨缺损的效果,探讨其作用机制。 方法 将IGF-Ⅱ,bFGF和bBMP,分别与PLGA复合成具有生物活性的三种诱导骨再生膜,用其修复兔桡骨骨缺损,以单纯PLGA膜作为对照组,分别在2、4、8周进行大体、组织形态学、四环素示踪和组织形态计量学观察骨缺损的修复情况,并作统计学分析;免疫组织化学检测三种生长因子存在时间。 结果 提取的小牛骨bBMP能诱导成骨,三种诱导骨再生膜均能在体内诱导新骨的形成,明显优于PLGA(对照组),其诱导效果呈现PLGA/bBMP(B组)

Construction of Three Kinds of Membrane Guided Bone Regeneration and Comparison of Their Osteogenesis Capability in Vivo

Abstract:Objective To construct three kinds of artificial periosteum with PLGA/bBMP(bovine bone morpho-genetic protein),PLGA/bBMP+IGF-Ⅱ(Isulin-like Growth Factor-Ⅱ),PLGA/bBMP+IGF-Ⅱ+bBFGF(fibroblast growth factor-2)and compare their effects of inductive ostegogenesis in repairing the rabbit′s radius defect,so as to study the mechanism.Methods The Artificial Periosteums were constructed by attaching bBMP、IGF-Ⅱand b FGF on the PLGA membrane and were implanted into the defects of radius in18rabbits models,gross,histological and im-munohistochemical examination was studied and tetracycline labelling,metrical histological analysis were studied at2,4,8,weeks.Results The bBMP could induce bone regeneration in muscle and three kinds of artificial periosteum was superior to the control group(A组),The osteogenic ability of three kinds of artificial periosteum was superior to the control group(A组),The osteogenic ability of three kinds of Artificial Periosteum was PLGA/bBMP(B)
