摘要:目的 从8910、TC1、SKOV3、CAOV和3AO卵巢癌细胞系培养上清液中获得外来体。方法 分别培养8910、TC1、SKOV3、CAOV和3AO卵巢癌细胞系,收集上清液,通过多步离心和超滤等方法分离出外来体。电镜下进行形态学观察。结果 8910和3AO细胞系培养上清液中分离出外来体,置于透射电子显微镜下观察,可见具有明显异质性的圆形或椭圆形小囊泡,直径约30-90nm,有完整的包膜。TC1、SKOV3和CAOV细胞系培养上清液中未分离出外来体。结论 8910和3AO卵巢癌细胞系可分泌外来体,两种细胞系来源的外来体在形态和大小上无明显差别,有可能作为卵巢癌免疫治疗的潜在抗原。

Obtainment and morphologic characteristics of exosomes acquired from ovarian cancer lines

ABSTRACT: Objective To obtain exosomes from the cell culture supernatants of 8910, TC1, SKOV3, CAOV and 3AO ovarian cancer cell lines. Methods 8910, TC1, SKOV3, CAOV and 3AO ovarian cancer cell lines were cultured separately and the cell culture supernatants were collected to seperate exosomes with multiple centrifugation and ultrafiltration. Morphology of exosomes were observed with electron microscopy. Results Exosomes were purified in 8910 and 3AO cell culture supernatants. Exosoems exhibited heterogenous round or elliptic microvesicles ranging from 30 to 90 nm in diameter with intact membrane. Exosomes were not purified in TC1, SKOV3, and CAOV cell culture supernatants. Conclusion 8910 and 3AO cell lines may secrete exosomes, which are potential specific antigens for immunotherapy against ovarian cancer cells.
