成鼠胸髓损伤后延迟植入雪旺细胞与胚胎脊髓细胞混悬液

  【摘要】目的 了解单纯雪旺细胞(schwanncell,SC)移植或附加胚胎脊髓细胞(fetal spinal cord cell,FSC)对中枢轴索再生的作用。方法 SC悬液(schwann cell suspension,SCS)单独和与FSC悬液(fetal spinal cord cell suspension,FSCS)联合植入成鼠胸段(T7)打击伤1周脊髓空腔中,相同量的Dulbecco改良细胞培养液(dulbecco s modified eagle medium,DMEM)植入作为对照。移植后2、6周进行HE、嗜银染色组织学观察,免疫组化髓鞘碱性蛋白(myelin basic protein antibody,MBP)染色观察SC及髓鞘,5-羟色胺(5-histamine antibody,5-HT)染色观察下行运动纤维;神经丝(neurofilament antibody,NF)染色观察FSC向神经元的分化。结果 在宿主中SC存活、增殖,FSC分化趋向成熟神经元。含SC的移植物中均见再生长入的宿主轴索,且多与束状排列的SC相伴随,二者MBP染色均为阳性。混合移植物中,5-HT阳性纤维相对丰富,对照组未见再生轴索。结论 自成鼠周围神经培养的SCS移植可促进脊髓轴索再生,形成髓鞘。SCS与FSCS联合移植可诱发部分脑干神经元轴索再生长入成鼠胸段脊髓。

Delayed Transplantation of Mixed Cultured Schwann Cells and Fetal Spinal Cord Suspension Into Injured Thoracic Spinal Cord of Adult Rat

  【Abstract】 Objective To observe the effect of Schwann cells suspension (SCS) grafts either alone or mixed with fetal spinal cord cells suspension (FSCS) on axonal regeneration of central nervous system. Methods The SCS and SCS+FSCS were implanted into the empty space left following injured thoracic (T 7) spinal cord injury in adult rats one week later, while the vehicle solution-DMEM was grafted in the controls. At 2 and 6 weeks post-transplantation, the samples were procured and studied histologically with silver and HE stains. In immunohistochemical assay, myelin basic protein antibody (MBP)was used to identify Schwann cells (SC) and myelin, serotonin antibody (5-HT), a specific neurotransmitter to raphe-derived axons, was used for descending motor fibers, and neurofilament antibody (NF) was used for the immature neurons derived from grafts.Results SC could survive and proliferate, FSC could differentiate in the host spinal cord, regrowing axons in grafts were associated with bundles of SCs, both axons and SCs were i
