  摘要 目的和方法:在大鼠主动脉球囊内皮剥脱模型上,于术后3、10、21 d观察在体和体外孵育血小板活化及应用血小板膜糖蛋白(GP)Ⅱb/Ⅲa受体拮抗剂精-甘-天冬-丝氨酸(RGDS)肽对其活化的影响。结果:球囊剥脱术后大鼠的血小板聚集明显增强,蛋白激酶C(PKC)活性明显增高,环磷酸腺苷(cAMP)活性明显降低,血小板膜活化的GPⅡb/Ⅲa受体密度明显增高,均以3 d组最为明显。应用RGDS肽体外孵育,可明显逆转上述改变。结论:血小板膜活化的GPⅡb/Ⅲa受体的变化,可能是内皮剥脱时介导血小板活化的关键途径。

Effect of Arg-Gly-Asp-Ser(RGDS) peptide on platelet activation after balloon angioplasty in rat aorta

  Abstract AIM and METHOD:The present study observed the effect of balloon angioplasty and RGDS peptide,the antagonist of platelet glycoprotein Ⅱb/Ⅲa receptor,on platelet activation in vivo and in vitro at 3,10 and 21 days after endothelial denudation with balloon.RESULTS: Platelet aggregation,protein kinase C(PKC) activity and platelet GPⅡb/Ⅲa receptor density are obviously enhanced, while cyclic monophosphate(cAMP) activity is decreased after balloon angioplasty.The most prominent changes occurred on the third day.When platelets were incubated together with 100μmol/L RGDS peptide it was found that RGDS peptide obviously counteracted the above changes.CONCLUSION:The changes of platelet GPⅡb/Ⅲa receptor might be the key pathway of endothelial denudation on platelet stimulation.

  经皮冠状动脉腔内成形术(percutaneous transluminal coronary angioplasty, PTCA)是冠心病治疗的有效措施,但病人术后可能发生血栓形成和血管再狭窄等并发症,其发生机理尚未完全阐明。临床和实验资料表明,活化血小板起了关键作用[1],它不仅能触发凝血过程,而且可以通过释放凝血酶、血栓素和生长因子等介质,促使新生内膜增殖,发生再狭窄。Tcheng等报道[2]应用血小板膜的纤维蛋白原(fibrinogen,Fg)结合受体-糖蛋白(glycoprotein,GP)Ⅱb/Ⅲa受体的单克隆抗体,临床实验有效防治PTCA术后再狭窄的发生。但是受损伤的血管激活血小板及阻断GPⅡb/Ⅲa受体对其活化影响细胞分子机制,目前尚不清楚。本工作在大鼠主动脉球囊内皮剥脱模型上,观察术后不同时期的血小板在体和体外孵育的血小板活化和细胞蛋白激酶C(protein kinase C,PKC)活性的变化,并观察应用血小板膜GPⅡb/Ⅲa受体拮抗剂精-苷-天冬-丝氨酸(Arg-Gly-Asp
