大鼠弥漫性脑损伤后MMP 9和GABA表达变化的研究 |

【摘要】 目的 探讨弥漫性脑损伤(DBI)后损伤区脑组织基质金属蛋白酶_9(MMP_9)和γ_氨基丁酸(GABA)的表达。方法 采用自由落体撞击法制作成年雄性SD大鼠弥漫性脑损伤模型,大鼠随机分为对照组、伤后3、6、12、24、72h和7d组。采用免疫组化ABC法测定MMP_9和GABA的表达。结果 MMP_9在DBI后表达逐渐增强,至伤后24h达到高峰,伤后7d仍保持较高水平;而GABA在DBI后表达逐渐减弱,至伤后12h达到低谷,伤后72h恢复正常。MMP_9和GABA表达时相和强度呈负相关关系。结论 DBI可引起损伤区脑组织MMP_9表达增强和GABA的表达减弱,可能在脑组织的继发性损害中起重要作用。

[Abstract] Objective The aim of this study was to investigate the expression of metalloproteinase_9 (MMP_9) and gamma_aminobutyric acid (GABA) in injured brain and to explore potential role of MMP_9 and GABA after diffuse brain injury (DBI). Methods The trauma was produced by a free falling weight on the exposed dura of right parietal lobe.The rats were randomly divided into control group and DBI groups at 3, 6, 12, 24 and 72h, and on 7d. The expression of MMP9 and GABA was examined by ABC immunohistochemistry.
