白细胞介素2基因转导对卵巢癌细胞系SKOV3免疫原性的影响

  【摘要】 目的 探讨白细胞介素2 (IL-2)基因转导对卵巢癌细胞系SKOV3免疫原性的影响。方法 应用流式细胞术,测定SKOV3细胞组织相容性白细胞抗原(HLA)HLA-ABC、HLA-DQ及HLA-DR分子的表达,51Cr-4h释放实验显示SKOV3、SKOV3/Neo、SKOV3/IL-2细胞对自然杀伤(NK)细胞和淋巴因子激活杀伤(LAK)细胞杀伤的敏感性。结果 SKOV3/IL-2细胞中HLA-ABC、 HLA-DQ 及HLA-DR分子的表达率(17.7%、19.1%及9.4%)较SKOV3(1.5%、2.4%及4.7%)和SKOV3/Neo(4.3%、6.9%及2.1%)显著提高;SKOV3、SKOV3/Neo和SKOV3/IL-2细胞对NK细胞的杀伤均不敏感,但SKOV3/IL-2细胞对LAK细胞的杀伤敏感性较SKOV3、SKOV3/Neo细胞提高约1倍,差异有非常显著性;肿瘤细胞与淋巴细胞混合培养显示,淋巴细胞有向SKOV3/IL-2细胞聚集的趋势。结论 IL-2基因转导可提高卵巢癌细胞系SKOV3的免疫原性。

Effects of Human Interleukin-2 Gene Transfer on the Immunity of Ovarian Cancer Cell Line SKOV3

  【Abstract】 Objective To study the effect of interleukin-2 (IL-2) gene transfer on the immunity of ovarian cancer cell line SKOV3.  Methods The expression of main histologic complex (MHC) molecular HLA-ABC, HLA-DR and HLA-DQ of SKOV3, SKOV3/Neo and SKOV3/IL-2 cells were detected by flow cytometry. The 51 Cr-4hr releasing test was employed to detect the killing effects of natural killer (NK) and lymphokine-activated killer (LAK) cells. Meanwhile, the tumor cell and lymphocyte co-culture was performed. Results The expression of HLA-ABC, HLA-DR and HLA-DQ molecular of hIL-2 gene modified cells was upregulated markedly. The 51Cr-4hr releasing test showed that the parental and gene modified cells were markedly resistant to NK cells, but the IL-2 gene modified cells was more sensitive to LAK cells. There were significant differences between them (P<0.01). The “cross-talk” of tumor cell with lymphocyte was observed when the hIL-2 gene modified SKOV3 cells were cocultured with lymphocyte. Conclusion IL-2 gene transfer can enhance the immunity of ovarian cancer cell line SKOV3.
