【摘要】 目的:观察苦参素对四氯化碳(CCl4)诱导的实验性肝纤维化的干预作用。方法:将SD大鼠分成正常对照组、模型组、苦参素小剂量治疗组(40 mg/kg)、中剂量治疗组(80 mg/kg)、大剂量治疗组(160 mg/kg),造模4周后苦参素治疗至实验第10周,取血测相关血清学指标,计算肝脏指数,测定肝组织匀浆中HYP和MDA含量。结果:苦参素能降低肝纤维化大鼠血清丙转氨酶(ALT)、谷草转氨酶(AST)、胆汁酸(TBA)、γ-谷氨酰转移酶(GGT)和肝脏指数,同时能减少肝组织中羟脯氨酸(HYP)和MDA含量(P<0.05或P<0.01)。结论:苦参素具有抗CCl4诱导的实验性肝纤维化的作用。

Preventive effect of Kushenin on formation of experimental hepatic fibrosis in rats

[Abstract] Objective:To investigate the intervention effect of oxymatrine on experimental liver fibrosis induced by carbon tetrachloride(CCl4) in rats. Methods: Sprague-Dawley (SD) 50 rats were randomly divided into 5 groups: the normal control group, model control group without treatment, model with oxymatrine treatment of low dose(40 mg·kg-1), middle dose(80 mg·kg-1) and high dose group(160 mg·kg-1). The rat liver fibrosis model was prepared by administration of CCl4. In the development of fibrosis, oxymatrine was administered to rats from the 4th week to 10th week after CCl4 induction. Then rats were sacrificed, the serology related indicant in rat serum, liver index, content of HYP and MDA in the homogenate of hepatic tissue were measured. Results: Oxymatrine lower the level of ALT, AST,TBA,GGT in serum and liver index as well reduced the HYP and MDA content in liver tissues in liver fibrosis model rats. Conclusion: Oxymatrine has protective effect on CCl4-induced liver fibrosis in rats.
