糖基化终产物对人单核细胞EMMPRIN基因表达的影响

【摘要】  目的:探讨糖基化终产物(AGEs)对人单核细胞细胞外基质金属蛋白酶诱导因子(EMMPRIN)表达的影响。方法:在培养的人单核细胞株中分别加入不同浓度AGEs干预24 h和同一浓度AGEs干预不同的时间点,以1640培养基和相应浓度的牛血清白蛋白为对照组。用逆转录聚合酶链反应检测EMMPRIN mRNA的表达水平。结果:AGEs干预人单核细胞的EMMPRIN mRNA水平与对照组相比差异有统计学意义(P<0.05);且其表达呈时间和浓度依赖性降低(P<0.05)。结论:AGEs以时间及浓度依赖的方式减少人单核细胞EMMPRIN mRNA的表达。

Effect of advanced glycation end products on the gene expression of EMMPRIN in cultured

    human monocytes

    [Abstract]   Objective:To investigate the effects of advanced glycosylation end products (AGEs) on the expression of extracellular matrix metalloproteinase inducer(EMMPRIN) in cultured human monocytes. Methods:The cultured human monocytes were incubated with AGEs (50,100,200,400 mg·L-1) for 24 h or 200 mg·L-1 AGEs for 8、16、24 and 48 h respectively,which were incubated with 1640 medium and BSA as a control. The mRNA expression of EMMPRIN in human monocytes was analyzed by RT-PCR. Results:Compared with that in cells incubated with 1640 and BSA, the level of EMMPRIN mRNA in cells treated with AGEs decreased significantly in a concentration-dependent and time-dependent manner. Conclusion:The AGEs inhibit the expression of EMMPRIN in cultured human monocytes.
