九子参总皂甙药理研究

提要 研究九子参总皂甙对小鼠中枢神经系统的作用。光电法测定小鼠自发活动,戊巴比妥钠用于观察协同作用,戊四氮电休克法研究抗惊厥作用,热板法测定镇痛作用。结果表明九子参总甙属皂甙类,对中枢神经系统有抑制作用,可减少小鼠自发活动,协同戊巴比妥催眠并有一定的镇痛作用,无对抗戊四氮和电休克惊厥作用,还能提高小鼠的耐缺氧能力,小鼠灌胃的LD50为1.65g/kg。

Pharmacological Study of Total Saponin Extracted from Silene Aprica

Abstract To investigate the central nervous effects of total saponin (TG) of Silene aprica in mice, methods of photocells technique for measurement of spontaneous activity, pentobarbital sodium for observation of sedation, anticonvulsant for study of anticonvulsant activities, were tested against convulsions induced by cardiazol and electroshock activities, hot plate method was for analgesic activity were used. Results: TG was belong to saponins and had depressive activities to central nervous system.TG inhibited significantly spontaneous activity in mice.TG was revealed to possess analgesic effect in mice to some extent and potentiated hypnotic activity of sodium pentobarbital at subthreshold dose. It was found that TG prolonged the survival time in anoxic mice and LD50 in mice(PO) was 1.65g/kg.

  九子参为石竹科植物女娄菜Silene aprica Turcz.ex Fisch,ef Mey. 〔Melandrium apricum (Turcz.)Rohrb.〕的全草,亦称女娄菜和罐罐花,在我国分布较广,西南各省均有分布。文献记载,九子参味苦、甘、平,主治体虚浮肿、痈肿和肿痛等证[1,2],在云南大理地区应用普遍。九子参所含成份未见报道,云南省药物研究所从中提得总甙。本文报道九子参的某些药理作用。

1 实验材料
1.1 药物:九子参总皂甙由云南省药物研究所提供,为淡黄色粉末,纯度为98%,易溶于水。
1.2 动物:昆明种小鼠由本校动物房繁殖,体重18~22 g。热板法镇痛用雄鼠,电休克用雄鼠,其它实验雌雄兼用。家兔由本校动物室提供,体重2.5 kg。
1.3 仪器:GJ-790I型三光道活动记数仪(上海光学仪器厂)。

2 方法和结果
2.1 对中枢神经系统的作用[3,4]
2.1.1对小鼠自发活动的影响:小鼠120只,均分成15组。阴性对照组5组(生理盐水0.2 ml/10g,口服),实验组5组(九子参总甙200 mg/kg,口服),阳性对照组5组(左旋四氢巴马汀L-THP400 mg/kg,口服)。分别于给药后15、30、60、120和240 min用GJ-790I型三光道活动记数仪记录小鼠在5
