党参对鼠J 774巨噬细胞吞噬活性的增强效应

摘要:目的:验证党参对机体免疫功能的增强效应并探索其作用机理。方法:用党参水提液加入鼠J 774巨噬细胞培养液中作微量细胞培养,以自动微量荧光分析仪直接检测其对细胞吞噬活性的影响。结果:显示党参水提液对巨噬细胞的吞噬活性有明显的增强作用,在浓度500~3 000 μg/ml时,与无药对照组(O)比较都有显著性差异(P<0.05),分别增强1.33~1.70倍。结论:巨噬细胞是重要的免疫活性细胞之一,对机体免疫反应的各个环节都起关键性作用,党参能增强机体免疫的效应,可能是通过其增强巨噬细胞的免疫活性而实现的。

The Enhancing Effect of Chinese Medicine Radix pilosulae on J 774 Macrophage

Abstract:Objective: To probe into the mechanism of the enhancing effect of Chinese medicine Radix Codonopsis (CR) on organic immune activity. Method: The murine macrophage J 774 cell line was chosen to cultured with CR aqueous extract in different concentrations and then, the phagocytic activity was measured directly using a computerized microplate fluorometer. Result: The phagocytosis of macrophage J 774 was enhanced significantly by CR solution in concentrations 500 μg/ml to 3 000 μg/ml (P<0.05). The peak value was at 2 000 μg/ml, which increased 170% compared with the control group. Conclusion: It is suggested that the mechanism of the enhancing effect of CR on organism is accomplished probably through the augment of macrophage function.

  党参(Radix Codonopsis, CR)为桔梗科党参属植物,干燥根入药,可补气养血、增强机体免疫功能。中医用于“阴虚气弱、气虚”诸证,可以代人参,惟作用较弱[1]。为探究党参增强机体免疫功能的作用机理,我们用党参水抽提液加入鼠J 774巨噬细胞培养液中,观察其对该巨噬细胞吞噬活性的影响。现将结果报道于下。
1 材料与方法
1.1 药物来源及制备:实验用党参购自西安市药材公司,陕西产。原药烘干,磨细粉。称取干粉10 g,加蒸馏水100 ml,置56℃水浴箱温浴1 h,悬液经2 000 r/min离心15 min,上清液经0.45 μm醋酸纤维滤器(Corning Glass Works, Corning, NY, USA)过滤消毒。4℃冰箱保存备用。
