肾癌细胞c

  摘要 目的:阐明c-myc表达在肾癌发生、发展中的作用及临床意义。方法:利用免疫组织化学ABC法结合微波抗原修复和流式细胞技术,对c-myc蛋白在52例肾癌组织及14例癌旁正常肾组织中的表达进行了定性、定量的测定。结果:肾癌组织中c-myc蛋白的阳性率和阳性细胞率分别为76.92%和65.45%±29.19%,高于正常肾组织的14.29%和8.59%±3.66%(P<0.005)。c-myc在肾癌组织中55.77%分布于胞浆,21.15%分布于胞核,而在正常肾组织则全部分布于胞核。其胞浆表达在病理G2、G3级高于G1级(χ2=9.244,P<0.005);临床分期T3、T4期高于T1、T2期(P<0.001)。结论:c-myc的胞浆表达促进肾癌的发生、发展,并与恶性程度、临床分期有关,是预后不良的重要指征。

Cellular localization of c-myc product and clinical
significance in renal cell carcinoma

  Abstract Purpose:To get to know the effect of c-myc protein on the onset and development of renal cell carcinoma and clinical progress.Methods:We have detected expression of c-myc protein qualitatively and quantitatively in 14 cases normal tissue and 52 cases renal cell carcinoma(RCC). Using immunohistochemical technique,and cytoflowmetry and cytoimmunofluorescence.Results:Normal tissue showed staining only of some nuclei while in RCC staining varied from predominantly nuclear to focal or pancytoplasma. Positive rate and positive cell rate of c-myc protein were 76.92% and 65.45%±29.19% respectively in RCC,they were significant higher than those (14.29% and 8.59%±3.66%) of the normal tissue (P<0.005). Cytoplasmic expression of c-myc was related to high pathological grade (χ2=9.244,P<0.005) and high clinical stage (P<0.001).Conclusions:Cytoplasmic expression of c-myc might play important roles in the pathogenisis and was helpful in the evaluation of prognose of patients in RCC.
