  摘 要 目的和方法:观察百日咳菌液诱导的感染性脑水肿不同时间大鼠脑含水量和伊文思兰(EB)含量的变化,以及MK-801预处理和尼莫地平(nimodipine)对感染性脑水肿脑含水量和EB含量的影响。结果:感染性脑水肿在注菌后30min,脑含水量和EB含量已明显升高,应用MK-801预处理或nimodipine治疗后,脑组织含水量和EB含量显著降低,脑水肿明显减轻。

The effect of nimodipine and MK-801 on brain edema induced
by injection of Pertusis Bacilli in rats

  Abstract AIM and METHODS:TO study the changes of brain water content and Evan blues (EB)in 82 SD rats.MK-801(0.5 mg.kg-1.d-1 in total of 2 injections) was given at 24 and 48 hours before the injection of Pertussis Bacilli(PB).Nimodipine(Nim) was also employed to show its effect on the brain edema induced by PB.RESULTS:The brain water content and EB content increased significantly after injection of PB(P<0.05), and that there was close relationship between water content and EB content. Nimodipine and MK-801 could significantly reduce brain water and EB content, and brain edema was also attenuated.Nimodipine and MK-801 have the protective effect on brain edema induced by PB.

  MK-801是N-甲基-D-天冬氨酸(NMDA)受体的非竟争性拮抗剂,最早作为抗癫痫和抗焦虑药,80年代发现其具有抗脑损伤和脑水肿作用,其作用与特异性阻断NMDA受体离子通道内部的结合位点、进而阻断钙离子内流有关[1],关于MK-801对感染性脑水肿的作用尚未见有文献报道;将尼莫地平(nimodipine)应用于感染性脑水肿及其作用机制的研究,文献很少有报道。为此,本研究采用感染性脑水肿模型,研究MK-801和特异性L-型钙离子通道阻断剂nimodipine对感染性脑水肿脑含水量和EB含量的影响,旨在为临床治疗提供实验和理论依据。

材料与方法
  1.动物分组:健康SD大鼠(符合国家二级动物标准)共82只,雌雄不分,随机分为4组,即(1)生理盐水对照(NS)组,19只;(2)百日咳菌液(PB)组(百日咳菌液由北京生物制品研究所提供的灭活菌液),25只;(3)Nimodipine治疗(Nim)组,20只;(4)MK-801预处理(MK-801)组,18只。各组又分为30min、4h和24h3个时间亚组。
  2.感染性脑水肿模型复制:经左颈内动脉于30s内匀速注入百日咳菌液0.2mL/kg(含菌量108亿/L)。注菌后5min内向腹腔内注入nimodipine (Nim治疗组,0.25mg/kg体重),24h nimodipine治疗组每8h重复注射1次相同剂量;MK-801预处理组于实验前48 h和24h向腹腔内注入MK-801
