骨碎补总黄酮对实验性兔膝骨性关节炎软骨及滑膜的影响

【摘要】 目的 研究骨碎补总黄酮对兔膝骨性关节炎的治疗效果,探讨其防治骨性关节炎的可行性。方法 32只新西兰大白兔随机分为正常对照组、模型对照组、骨碎补总黄酮组和维固力组。以关节制动法建立兔膝骨性关节炎模型。成模后灌胃给药,8周后镜下观察骨碎补总黄酮对动物模型关节软骨、滑膜组织形态学及基质金属蛋白酶-3表达的影响。结果 骨碎补总黄酮组与模型对照组比较滑膜及软骨组织形态学改变明显较轻(P<0.01),滑膜、软骨组织中基质金属蛋白酶-3表达减少(P<0.01);骨碎补总黄酮组与维固力组相比,各指标间差异无统计学意义(P>0.05)。结论 骨碎补总黄酮能够减轻兔膝骨性关节炎的软骨退行性变和滑膜炎症,其作用机制可能与调节基质金属蛋白酶-3表达有关。

  Effect of osteopractic total flavone on cartilage and synovia

  of knees with osteoarthritis in rabbits

  Abstract:Objective To observe the effects of osteopractic total flavone (OTF) on treatment of osteoarthritis in experimental rabbits and to explore the feasibility of using OTF in the prevention and treatment of osteoarthritis.Methods 32 rabbits were randomly divided into normal group, model control group, OTF group and Glucosamine Sulfate Capsules group. The rabbit osteoarthritis model was established by immobilizing the knees of rabbit, followed by gastrogavage. Four weeks later, observe the effects of OTF on the histology of joint cartilage, synovial and the levels of matrix metalloproproteinases (MMP)-3 in cartilage and synovial were observed.Results The histological changes of cartilage and synovia were lower in OTF group than those in model group (P<0.01). The levels of MMP-3 in cartilage and synovia decreased. There were significant differences in OTF group and the model group (P<0.01), while the differences in OTF group and Glucosamine Sulfate Capsules group were insignificant (P>0.05).Conclusion OTF can reduce the degenerative changes of joint cartilage and synovitis in osteoarthritis model, of which the mechanism is related to the adjustment of the levels of MMP-3.
