淫羊藿总黄酮对人工关节磨损微粒引起单核细胞分泌溶骨性因子的影响

【摘要】 目的 探讨淫羊藿总黄酮防治人工关节松动的可行性。方法 采集30例人体外周血,分离单核细胞,分组培养。实验分4组:A组(对照组),仅单核细胞;B组,单核细胞及超高分子聚乙烯微粒;C组,单核细胞、超高分子聚乙烯微粒及帕米磷酸钠(10 mg/L);D组,单核细胞、超高分子聚乙烯微粒及淫羊藿总黄酮(10 mg/L)。培养48 h后,检测细胞上清液中肿瘤坏死因子-α(TNF-α)、白细胞介素-1(IL-1)和白细胞介素-6(IL-6)的含量。结果 B组分别明显高于A、C、D组(P<0.01);D组与C组之间的差异无统计学意义(P>0.05)。结论 淫羊藿总黄酮可有效抑制由于超高分子聚乙烯微粒刺激单核细胞而分泌的溶骨性因子TNF-α、IL-1和IL-6。淫羊藿总黄酮的效果与帕米磷酸钠相似,它有望成为将来防治人工关节无菌性松动的一种很有潜力的药物。

  Effect of total flavones of Epimedium on the secretion of osteolytic factors

  from human monocytes induced by wear particles

  Abstract: Objective To explore the rationale of using total flavones of Epimedium (TFE) to prevent and treat aseptic loosening of prothesis. Methods Blood samples obtained from 30 healthy human donors were processed to get the monocytes to perform the present experiment in 4 groups: Group A, contining monocytes only; Group B, monocytes+ultrahigh molecular weight polylethylene(UHMWPE)debris; Group C, monocytes+UHMWPE debris+pamidronate(10 mg/L); Group D: monocytes+UHMWPE debris+TFE(10 mg/L). After 48 hours of incubation, the mixtures were tested for osteolytic factors including TNF-α, IL-1 and IL-6. Results The levels of these factors were much higher in group B than in the other 3 groups. There was no significant difference between group C and group D (P>0.05). Conclusion TFE can effectively inhibit the UHMWPE-stimulated secretions of TNF-α, IL-1 and IL-6 by monocytes. It is quite likely for RFE to become a potential therapeutic candidate for the treatment and prevention of aseptic loosening.

  人工关节磨损微粒能刺激溶骨性因子的分泌而引起假体周围骨溶解,最终导致人工关节无菌性松动[1~3]。本实验采用人工关节松动的体外生物学模型研究淫羊藿总黄酮对溶骨性因子分泌的影响,并与已用于临床研究防治人工关节松动的二磷酸盐类药物比较,探讨淫羊藿总黄酮防治人工关节松动的可行性。
