花椒油素对肺血栓及6

摘要:以胶原和肾上腺素混合液给小鼠,制造肺血栓模型,用放免法测定6-酮-前列腺素F1α(6-keto-PGF1α)和血栓素B2(TXB2)含量。结果表明,花椒油素(Xanthoxylin,XT)1.0mg/kg,2.0mg/kg,4.0mg/kg和8.0mg/kg使小鼠的存活率增加28.6%~71.2%;使6-keto-PGF1α的产量提高56.3%~210.8%,使TXB2的产量降低21.6%~56.8%。6-keto-TGF1α与TXB2的比值明显提高,对照组为1.15,药物组为2.29,3.13,5.53,8.26。体外实验证明,XT0.037μmol/L,0.185μmol/L,0.924μmol/L,9.240μmol/L显著地抑制血小板聚集和TXB2产生,前者的抑制率为22.7%~55.3%,后者为20.5%~50.7%;但同时增加6-keto-PGF1α的产量,提高率为27.3%~47.0%。6-keto-PGF1α与TXB2比值:对照组为1.23,药物组为2.07,2.44,3.28,3.85。

Effects of Xanthoxylin on the Amount of 6-keto-prostaglandin Fla and
Thromboxane B2 in Mice with Pulmonary Thromboembolism

Abstract:Model mice with pulmonary thromboembolism were established by administration of mixture of collagen and adrenaline.Radioimmunoassay was used to detect the serum level of 6-keto-prostaglandinFla(6-keto-PGFla) and thromboxane B2(TXB2).Results showed that xanthoxylin being at the concentration of 1.0,2.0,4.0,8.0mg/kg,the survival rate of mice was increased by 28.6%~71.2%,the amount of 6-keto-PGFla by 56.3%~210.8%,and the amount of TXB2 by 21.6%~56.8%.The ratio of 6-keto-PGFla and TXB2 was enhanced markedly,being 1.15 in the control group, and 2.29,3.13,5.53 and 8.26 in the medication groups respectively.In vitro,xanthoxylin in the dosages of 0.037,0.185,0.924,9.240 μmol/L significantly inhibited the aggregation of platelet and the production of TXB2,with the inhibition rate of 22.7%~55.3%
