实验性大鼠胚胎脊髓移植对损伤脊髓影响的观察

  【摘要】目的 建立了一个胚胎脊髓(fetal spinal cord)组织的低温保存方法和一个损伤移植模式。方法 通过病理学、组织化学和免疫组织化学的研究,选用胎龄14天的Wistar大鼠,分离胚胎脊髓,逐步降温,最后置于液氮中保存。15天后取出,快速复温,并植入损伤大鼠脊髓内。全部动物于伤后8周处死。结果 经过低温保存的移植物能够在异体脊髓内存活并与宿主组织融合,其在损伤移植区的胶质细胞增生少于对照组。移植组宿主再生神经纤维能穿越宿主—移植物界面进入移植区。结论 经冰冻保存的胚胎脊髓移植物改变了宿主的再生环境,并显示对损伤轴突有营救作用。

Experimental Transplantation of Fetal Spinal Cord and Observation of Its Influence to the Injured Spinal Cord in Rat

  【Abstract】 Objective To establish a method of cryopreservation of fetal spinal cord (FSC) tissue and a model of spinal cord injury for the transplantation.Methods FSC tissue was harvested form E14 Wistar rats and cut into slices of 1 cubic mm. It was put into protective liquid for cryopreservation in liquid nitrogen( -195.8 ℃ ).Spinal cord injury(SCI) model was produced in 24 wistar rats at T 7 level using modified Allen’s impact method with 50 g.cm .The animals were divided into transplanted and control group randomly. After 4 weeks, cryopreserved FSC was grafted into injured segments in the transplanted group. All animals were sacrificed at 8 weeks following the impact injury.The pathologic, histochemical and immunohistochemical changes of the sample were observed.Results The study showed that the grafts of FSC might survive in allogenic spinal cord and fused with the host neuron, passed through the interface and extended into transplanted area. In the control group, the regenerated fibers were blocked.Conclusion This finding suggested that cryopreserved FSC tissue transplantation was able to change the regeneration environment of the host, and ingrowth of axons of host neurons into the transplanted area played the role of salvage of the injured axons.
